Why did the uridine content increase in my sequence?

Issue

The uridine content increased instead of decreased after codon optimization.

Cause

Uridine depletion will reduce some, but when combining both organism and uridine depletion parameters, the uridine content in the optimized sequence might still be higher than the starting material. The optimizer will try its best to deplete uridine, but it can only decrease the uridine level so much without jeopardizing the low GC content. Thus, uridine level may actually increase with respect to the original sequence. 

Resolution

To obtain an optimized sequence with reduced uridine content, set the organism to None to select only the Uridine depletion parameter.
