DNA and RNA contain many components, called parts. Using the Registry, scientists can track and share parts across teams. For example, an mRNA sequence could have parts designating the 5’UTR, exons, and 3’UTR. Scientists working with plasmids could:
-
Define and track plasmid components
-
Intelligently autofill plasmids with annotations and their components
-
Search for shared plasmids and filter by gene, promoter, tag, etc.
Creating parts
Before configuring part links, you need to create parts. You can create parts manually and from existing sequences, or by importing them.
Create parts manually
To enter parts manually:
-
Click + in the left-side menu, hover over DNA/RNA Sequence, and select New DNA/RNA Sequence.
-
Enter the sequence name and select the sequence type and topology.
-
Enter the bases, then click Create.
-
In the Metadata tab, select a schema, then click Register.
-
In the modal, select the name and Registry ID settings from the drop-down menu, then click Register.
-
Register the sequence and choose the correct entity type. For example, promoter, terminator, 5’ cap, etc. After it’s registered, you can automatically find and link the part in other sequences.
Create parts from an existing sequence
-
Open the DNA or RNA sequence that contains the part.
-
Select the bases then click Create in the right-side menu above the sequence view.
-
Select New Part, then enter the part name and select a folder to store it in.
-
Register the part and select the entity type.
Configure part links
To configure part links:
-
Click your avatar in the bottom-left corner of your tenant, then under Feature Settings, select Registry Settings.
-
In the top-right corner of the Entity Schemas page, click Create.
-
Enter a schema prefix and name. We recommend using the name of the part you are storing. For example, Promoter, Enhancer, etc.
-
In the Entity Type drop-down menu, select DNA Sequence or RNA Sequence..
-
Add your parts, then click Next.
-
Search for collaborators and click Add Collaborators to add them to your list of collaborators.
-
Select each collaborator’s access policies from the drop-down menus in the Access policies column.
-
Click Create.
